I’m interested in making a custom fill pattern, similar to the tatami fill but with a long slow curve to it. Is there a method for regular users to do custom fill work?

I need to make a video on this.  You can play around with the tatami fill pattern by just opening it and changing the points make sure you name it something else.  The fill pattern has and extension called .filpat.  A fill pattern is just another graphic type.
